Unraveling the Charge Heterogeneity of Biotherapeutics: icIEF Fractionation for Mass Spectrometry Characterization

Imaged isoelectric focusing (icIEF) has been established as the standard technique for profiling charge heterogeneity, which is a critical quality attribute (CQA) of biotherapeutics. The in-depth characterization of individual charge variants - currently done by mass spectrometry - requires either collecting fractions of charge variants with traditional ion exchange chromatography (IEX) or by coupling the capillary electrophoresis (CE) directly to mass spectrometry (CE-MS).

 

In this webinar, we present a new and innovative MauriceFlex™ platform that can perform icIEF-based fractionation, in addition to routine CE analysis. The fractionation utilizes a special cartridge to perform isoelectric focusing to separate charge variants which are subsequently eluted via chemical mobilization for fraction collection. The MauriceFlex™ system provides a true pI-based fractionation method that is flexible and overcomes some of the deficiencies of IEX-based fractionation and direct CE-MS. The complete workflow from fractionation to mass spectrometry characterization will be illustrated.
