It is important to confirm the sequences and post-translational modifications (PTMs) of protein therapeutics to ensure drug safety and efficacy. Middle-down mass spectrometry (MS) combines the advantages of the bottom-up and top-down MS approaches by offering high sequence coverage of subunits following simple sample preparation. Traditionally, a middle-down workflow requires extensive method development and often involves multiple fragmentation techniques and/or injections to obtain high sequence coverage.
This webinar discusses a single injection workflow based on electron activated dissociation (EAD) that addresses these challenges in middle-down biotherapeutic characterization. It leverages the descriptive and reproducible EAD fragmentation provided by the ZenoTOF 7600 system with automated data analysis using Biologics Explorer software. As a result, the workflow significantly reduces the time and effort required for middle-down analysis of protein therapeutics, enabling routine middle-down analysis of monoclonal antibodies (mAbs) with consistently high sequence coverage.
