This talk introduces the Leica STELLARIS 8 τ-STED system where STED Nanoscopy meets Leica’s Fast Lifetime Contrast (FALCON) imaging to enable a new level of super resolution performance. Since the advent of super-resolution fluorescence microscopies, the characterization of macromolecular complexes and cellular structures at the sub-diffraction level has become a reality and STED Nanoscopy has become an accepted confocal-based technology to deliver resolutions in the tens of nm. τ-STED combines the optical signals from STED imaging with the photophysical information from the fluorescence lifetime at unprecedented speed and sensitivity. τ-STED uses phasor analysis in a novel way to enable increased STED resolution and elimination of uncorrelated background noise even with reduced excitation and depletion powers. This talk will also present some interesting application examples, including 2D and 3D STED in live and in fixed specimens, as well as for multicolor applications.
