Multi-wavelength analysis of hydrodynamic data collected on the latest analytical ultracentrifugation instruments provides additional resolution and insights into hetero-interacting systems by adding spectral characterization to the traditional hydrodynamic separation of different molecules present in a mixture.
In this talk Dr. Demeler will provide an introduction into this new methodology, its computational challenges, and explain its benefits for the investigation of biopolymer interactions in the solution phase. The technique is used to exploit different chromophores present in different chemicals interacting with each other in solution, such as proteins and nucleic acids, proteins with variable ratios of amide backbone absorbance compared to tryptophan/tyrosine absorbance, proteins linked to fluorescent proteins, or proteins containing unique chemical groups like metal-bound porphyrins, or fluorescent tags. Dr. Demeler will also illustrate this advance by presenting several examples from his laboratory, covering situations commonly encountered in the characterization of biomolecular interactions.
