As the SARS-CoV-2 virus that causes COVID-19 keeps mutating, there is a growing need to sequence the viral genome from many COVID-19-positive samples to better understand the genetic sequence of the virus. Identification of viral variants enables the development of diagnostic tests and therapeutics against the virus. Sequencing viral variants requires high- throughput sequencing methods. The ideal protocol would have a low-cost, high-throughput and simple workflow compatible with a wide variety of sequencing instruments. To address this need we discuss a method deployed on the Beckman Coulter Life Sciences tip-based and acoustic liquid handling platforms to process samples in batches of 384 samples. The protocol can be completed in a single shift and uses only three tips per sample.
