An unbiased characterization of the human microbiome is crucial to a more complete understanding of microbial effect on human health and diseases. The rapidly reducing costs of sequencing and development of specialized analysis pipelines have made whole metagenome sequencing an attractive option. However, for many microbiomes, the presence of large amounts of host nucleic acid content has meant that data from low biomass microbes is swamped by information of little interest to the researcher. A need exists for an unbiased and easily implemented method of host nucleic acid content depletion. To this end, we have developed a host depletion technology (CRISPRclean) that leverages the CRISPR-associated protein 9 (Cas9) and specifically designed guide RNAs to remove human nucleic acid fragments from human microbiome samples prior to sequencing.
This webinar will review the results of our preliminary in-vitro studies. By redistributing reads to lowly abundant or expressed molecules within the sequencing library, a greater taxonomic resolution is achieved. Jumpcode depletion technology helps you overcome the overwhelming human host contamination and thus maximizes microbial genome coverage and gain higher taxonomic resolution.
